چکیده

plement in the presence of Ca2+, but absenco of Mg2+, certain components (Cx') of complement combine with EA, while the other components (Cy') of complement are left in the fluid phase. In order to determine the complement components involved in EACx' and Cy', carp seru m was inactivated by carrageenin (Cn), ammonia (Am), hydrazine (Hd), zymosan (Z) and heat treatment (13), and each inactivated serum was combined with EACx'or (EA+Cy'). Consequently, the combinations of EACx'+Cn, EACx'+Am, and (EA+Cy')+Z resulted in hemolysis, while the other combinations were not hemolysed. This suggests that EACx' comprises two components which are destroyed by carrageenin and by ammonia, probably the Cl and C4 components, and that Cy' contains the compovents which are removed by zymosan. Furthermore, it was found that in the reaction between EA and Am, the first component combines with EA, with formation of EACl, and that this complex is hemolyzed by Cn. This confirms that the component inactivated by carrageenin corresponds to the Cl component of complement. From these data, it was concluded that the intermediate complex EACx'obtained in immune hemolysis by carp complement is a complex corresponding to mammalian EAC1, 4, and that Cy' is a mixture of the other components.

An Improved Method for the Preparation of EAC14, an Intermediate in Immune Hemolysis by Carp Complement

We have succeeded in preparing EAC14 (sheep erythrocytes sensitized with carp antibody and complement components, Cl and C4) by incubating EA with carp serum in the presence of 5 mM TTHA (triethylenetetramine-N,N,N’,N’:N”:N”’hexaacetic acid), a selective chelator of Mg’+, and 5 mM CaCl* at pH 7.4. The optimal conditions for preparing EAC14 were as follows: ionic strength, 0.085; reaction temper...

Interference with Immune Hemolysis by Glycoprotein Antigens*, by E. R. Arquilla,

Hemagglutination and hemolytic titrations of antisera using red cells to which antigens have been conjugated with bis(diazo)benzidine have been used as a measure of antibody activities (1--4) in this laboratory. Hemagglutination and immune hemolysis fitrations have comparable sensitivities. Immune hemolysis, however, can be modified to give a high degree of precision (5). Both methods have been...

STUDIES ON IMMUNE CELLULAR INJURY II. FUNCTIONAL ROLE OF Ctl ESTERASE IN IMMUNE CYTOTOXICITY* BY IRWIN

I t was demonstrated in the preceding paper (1) that incubation of primary isolates of normal human amnion cells with specific rabbit antibody and normal human serum resulted in immune cellular injury as measured by two independent criteria of cytotoxicity. I t was further shown that the factors in normal human serum which were required in addition to specific antibody for production of cytotox...

STUDIES ON IMMUNE HEMOLYSIS: PREPARATION OF A STABLE AND HIGHLY REACTIVE COMPLEX OF.SENSITIZED ERYTHRO- CYTES AND THE FIRST COMPONENT OF COMPLEMENT (EAC'I); INACTIVATION OF CELL-FIXED C'l BY SOME COMPLEMENT REAGENTS* BY PAUL G. KLEIN, M.D

The interaction of sensitized sheep erythrocytes (EA) with complement (C') leads to hemolysis of the red cells in a sequence of several reaction steps. Levine and Mayer (1) have established that the components C'I and C'4 first interact with the sensitized cells to form an intermediate complex EAC'1,4. Whether the transfer of hemolyticaUy active C'1 to sensitized erythrocytes is dependent upon ...

The S region of the H-2 complex controls the serum levels of two proteins, Ss and Sip, which are closely related antigenically (1-4). Several observations suggest that the Ss protein may be part of the complement system and homologous to C4. F(ab')2 fragments of antibody to Ss inhibit the hemolytic activity of mouse serum (5) and Ss antigenic determinants are incorporated into immune complexes ...
